3.5 HD SATA with USB adapter

Hi i have an 500gb sata 3.5 with case properly working on Configurable USB 
Loader, every time i try to use on Nintendont the program dont find my 
USB:/games folder, saying no FAT and directory found, on CFG Loader all 
gamecubes game show and also Wii games are playable, from Nintendont or CFG if 
i try to run game or program exit , wii power off , restart...
The USb case come with 12v adapter properly working and connected , no 
partitioned , formater on FAT32 with 32k cluster.
Solutions ?
Please tell me if i need change cluster, create another partition...
